| Abstract: Traditional methods of identifying components in a mixture normally involve isolation of the components of interest. If a component is labile, it may decompose or rearrange during the isolation procedure. Multiple preparatory chromatographic separations may be necessary to provide a sufficient quantity of sample for NMR analysis in a 5mm NMR tube. The coupling of an analytical HPLC and detector with an NMR was the first step in the development of hyphenated NMR technologies. LC-NMR analysis of components eliminates the need for isolation and NMR data may be acquired on many components within a single chromatographic run. The addition of a mass spectrometer to an LC-NMR system provides two primary functions; the acquisition of a mass spectrum of the individual components and the highly selective nature of mass spectroscopy is used as a discriminating factor for peaks prior to NMR analysis. MSn measurements are possible with the incorporation of a specialized LC-NMR-MS interface. A complete description of the LC-NMR and LC-NMR-MS hardware and working modes will be presented. | |||||||||
